Protein Description:Full Length of Mature Protein

Tag Info:Tag-Free

Mol. Weight:15.1 kDa

Biological_Activity:Fully biologically active when compared to standard. The ED50 as determined by a cell proliferation assay using rat C6 cells is less than 0.1 ng/ml, corresponding to a specific activity of ? 5.0 × 107 IU/mg.

Purity:>97% as determined by SDS-PAGE and HPLC.

Endotoxin:Less than 1.0 EU/µg as determined by LAL method.

Form:Lyophilized powder

Buffer:Lyophilized from a 0.2 m filtered 1 × PBS, pH 7.4, with 0.05 % Tween-20

Reconstitution:We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20?/-80?. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Notes:Repeated freezing and thawing is not recommended. Store working aliquots at 4? for up to one week.

Alternative Name/ Alias:hGDNF, ATF

Relevance:Neurotrophic factor that enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake. {ECO:0000269|PubMed:8493557}.

PubMed ID:8493557; 7867768; 15372022; 15489334; 10366742; 9811930; 8988018; 22081608; 9359036; 19478429; 8968758; 8896568; 8896569; 9497256; 10917288

Function:Neurotrophic factor that enhances survival and morphological differentiation of dopaminergic neurons and increases their high-affinity dopamine uptake.

Involvement in disease:Hirschsprung disease 3 (HSCR3); Congenital central hypoventilation syndrome (CCHS); Pheochromocytoma (PCC)

Subcellular Location:Secreted

Protein Families:TGF-beta family, GDNF subfamily

Tissue Specificity:In the brain, predominantly expressed in the striatum with highest levels in the caudate and lowest in the putamen. Isoform 2 is absent from most tissues except for low levels in intestine and kidney. Highest expression of isoform 3 is found in pancreatic islets. Isoform 5 is expressed at very low levels in putamen, nucleus accumbens, prefrontal cortex, amygdala, hypothalamus and intestine. Isoform 3 is up-regulated in the middle temporal gyrus of Alzheimer disease patients while isoform 2 shows no change.
