Alternative Name(s):Ikaros family zinc finger protein 1 (Lymphoid transcription factor LyF-1) (IK1) (IKAROS) (LYF1) (ZNFN1A1)

Relevance:Transcription regulator of hematopoietic cell differentiation. Binds gamma-satellite DNA. Plays a role in the development of lymphocytes, B- and T-cells. Binds and activates the enhancer of the CD3-delta gene. Repressor of the TDT gene during thymocyte differentiation. Regulates transcription through association with both HDAC-dependent and HDAC-independent complexes. Targets the 2 chromatin-remodeling complexes, NuRD and BAF, in a single complex, to the beta-globin locus in adult erythrocytes. Increases normal apoptosis in adult erythroid cells. Confers early temporal competence to retinal progenitor cells. Function is isoform-specific and is modulated by dominant-negative inactive isoforms.

Reference:"The Ikaros gene, a central regulator of lymphoid differentiation, fuses to the BCL6 gene as a result of t(3;7)(q27;p12) translocation in a patient with diffuse large B-cell lymphoma." Hosokawa Y., Maeda Y., Ichinohasama R., Miura I., Taniwaki M., Seto M. Blood 95:2719-2721(2000)

Purity:Greater than 85% as determined by SDS-PAGE.

Form:Liquid or Lyophilized powder

Buffer:If the delivery form is liquid, the default storage buffer is Tris/PBS-based buffer, 5%-50% glycerol. If the delivery form is lyophilized powder, the buffer before lyophilization is Tris/PBS-based buffer, 6% Trehalose, pH 8.0.

Reconstitution:We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20?/-80?. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Storage:The shelf life is related to many factors, storage state, buffer ingredients, storage temperature and the stability of the protein itself. Generally, the shelf life of liquid form is 6 months at -20?/-80?. The shelf life of lyophilized form is 12 months at -20?/-80?.

Notes:Repeated freezing and thawing is not recommended. Store working aliquots at 4? for up to one week.

Function:Transcription regulator of hematopoietic cell differentiation

Involvement in disease:Immunodeficiency, common variable, 13 (CVID13)

Subcellular Location:Nucleus, Note=In resting lymphocytes, distributed diffusely throughout the nucleus, Localizes to pericentromeric heterochromatin in proliferating cells, This localization requires DNA binding which is regulated by phosphorylation / dephosphorylation events, SUBCELLULAR LOCATION: Isoform Ik2: Nucleus

Protein Families:Ikaros C2H2-type zinc-finger protein family

Tissue Specificity:Abundantly expressed in thymus, spleen and peripheral blood Leukocytes and lymph nodes. Lower expression in bone marrow and small intestine.
