ClonExpress® II utilizes an Exnase II-mediated recombination system, allowing seamless fragment insertion by leveraging 15-20 bp homologous overlaps between the vector and insert. The process is simple:
- Design Overlaps: A short homologous sequence (15-20 bp) is added to the insert via PCR.
- Recombination Reaction: The linearized vector and PCR-amplified insert are mixed with Exnase II and incubated at 37°C for 30 minutes.
- Transformation: The reaction product is directly transformed into competent cells, yielding a true positive rate >95%.
This kit contains Exnase II and an optimized buffer with a unique recombinant enhancer that significantly improves recombination efficiency. Additionally, Exnase II is fully compatible with restriction digestion and PCR reaction systems, allowing direct use of digestion products and PCR inserts without purification, further simplifying the workflow.
